Deoxyribonucleic acid (DNA) extraction in abaca (Musa textilis Née), the source of strongest natural fiber (Manila hemp), is difficult due to its fibrous nature, high cellulose content, and polyphenol compounds. Thus an optimized DNA method required for extracting quality next-generation sequencing applications. This study, hence, aimed test compare five different methods molecular weight from leaves. The are traditional cetyl trimethylammonium bromide (CTAB) (Protocol 1), CTAB with polyvinylpyrrolidone (PVP) 2), 0.3 % ?-mercaptoethanol 3), sodium dodecyl sulfate 4), Triton X-100 PVP 5). use a throughput homogenizer (TissueLyserII) was also tested tandem select protocols applications high-throughput extraction. two varieties were extracted via Protocol 3 sent based on Illumina Novaseq platform. After passing control parameters library preparation, found be simplest most consistent average yield applications, while 4 determined have shortest processing time. Together TissueLyserII-facilitated homogenization, appropriate combination samples which will useful genotyping-by-sequencing (GBS) strategies as tool plant breeding.
